NAD+ and NMN: The Cellular Energy System Few People Talk About
You probably know that your energy declines as you age. You feel it in the gym, at work, in how long it takes to recover from a bad night of sleep. Most people chalk this up to "getting older" and leave it there. But there is a specific, measurable reason your cells produce less energy every year after 30, and it has a name: NAD+ decline.
If you care about how your body performs over the next decade, it's worth understanding what NAD+ does, why it drops, and what the research says about rebuilding it.
Your Cellular Battery Is Running Low
NAD+ (nicotinamide adenine dinucleotide) is a coenzyme found in every cell in your body. It is required for hundreds of biological processes, including energy production in the mitochondria, DNA repair, and the activation of sirtuins, a family of enzymes associated with longevity and cellular maintenance.
Think of NAD+ as the charge level on your cellular battery. When levels are high, your cells have the raw material to produce energy efficiently, repair damage, and maintain themselves. When levels are low, all of those processes slow down.
The problem for most people is that NAD+ levels decline significantly with age. This decline is associated with reduced mitochondrial function, changes in inflammatory markers, slower recovery, and many of the changes we typically attribute to aging. And it's not speculative anymore; NAD+ decline is well documented and its connection to cellular function is now generally recognized as established science. The question researchers are working on is what to do about it.
NMN: The Direct Route to NAD+
Your body can make NAD+ through several pathways, but the most direct route uses a precursor called NMN (nicotinamide mononucleotide). NMN is a naturally occurring form of vitamin B3 that gets converted to NAD+ after absorption. It is the last step before NAD+ in the biosynthetic pathway, which just means your body does not need to do very much "conversion" work to start using it.
The key question for any NAD+ precursor is simple: does taking it orally actually raise NAD+ levels in humans? For NMN, the answer appears to be yes:
- A 60-day double-blind, randomized controlled trial in 66 healthy adults aged 40 to 65 found that 300mg per day of NMN (our formula uses 500mg) raised blood NAD+/NADH levels by 38 percent from baseline. The placebo group saw a 14.3 percent increase, making the net advantage roughly 24 percentage points. Increases were already measurable at 30 days (Huang et al., 2022).
- A separate dose-response trial in 80 middle-aged adults tested three NMN doses over 60 days. All groups showed significant NAD+ increases, with the 300mg group rising from 11.8 to 32.6 nM (Yi et al., 2023). Higher doses showed even larger gains, though optimal dosing is still being established.
So the foundational mechanism works. Oral NMN gets absorbed, reaches your cells, and converts to NAD+. The next question is whether that NAD+ increase translates to outcomes you can feel.
What the Clinical Trials Show
The human research on NMN is still early, but the results so far are encouraging:
- A 12-week randomized, double-blind study in 108 older Japanese adults found that NMN supplementation improved lower limb function (measured by sit-to-stand tests, with an effect size of d=0.72) and reduced drowsiness, with the strongest effects from afternoon dosing (Kim et al., 2022).
- In healthy older men, chronic NMN supplementation elevated blood NAD+ levels and improved markers of muscle function, including gait speed and grip strength (Igarashi et al., 2022). Note: These improvements were nominally significant and need validation in larger studies, but they suggest the NAD+ elevation is translating to functional outcomes.
There are now six published human studies on NMN that we cite to on rbrnsupps.com. That is a meaningful body of evidence for a relatively new supplement ingredient, though it is still early compared to something like ashwagandha with over 140 studies. PQQ: Building New Engines
NAD+ is the fuel. But fuel is only half the equation. You also need engines to burn it.
Those engines are your mitochondria, the organelles inside every cell that convert nutrients into ATP, the energy currency your body runs on. You have trillions of them, and their number and efficiency decline with age just like NAD+ does.
PQQ (pyrroloquinoline quinone) addresses this side of the problem. It is a small quinone molecule shown in preclinical research to activate PGC-1alpha, the master regulator of mitochondrial biogenesis (in layman's terms, the process of building new mitochondria). (Bauerly et al., 2011).
The metaphor we use: NMN is like making sure you have plenty of high-quality fuel. PQQ is like building more engines to burn that fuel. Together, they address cellular energy from two different angles.
PQQ also functions as an unusually stable antioxidant. Most antioxidants are spent after a single reaction. PQQ can undergo thousands of REDOX cycles before being depleted, giving it a much longer working life inside your cells.
A human study found that dietary PQQ altered indicators of inflammation and mitochondrial-related metabolism in healthy subjects, providing early evidence that the mechanistic effects seen in cell studies translate to humans (Harris et al., 2013).
At RBRN, we're always honest about the limits what studies conclusively prove, and this article is no exception. Human clinical data on PQQ is very limited, just a handful of studies. For this one, most of the mechanistic research comes from cell culture and animal models. The mitochondrial biogenesis pathway is well understood and the preclinical data is compelling, but we do not yet have the depth of human evidence that exists for ingredients like NMN or ashwagandha.
We include PQQ at 20mg (the top of the 10 to 20mg range used in most available human research) because the underlying biology is sound, it complements NMN through a completely different pathway, and the safety profile is very good. We are transparent that this is an emerging ingredient where the science is still catching up to the promise. RBRN Vitality includes it based on mechanistic rationale and early data, not because the human research is settled. Anyone who tells you otherwise is lying to you.
